Q: Is it true that soon we will be able to radically change how we get older? — Frank I., Urbana, Ill.

A: I’m about 80% sure, based on the explosive growth of research into the mechanisms of aging, that a revolution in longevity is coming. Lab studies that prove this are impressive. So far scientists have engineered age reversal using 14 different approaches, each in at least two animal models.

And this is translating into significant research in humans. For example, researchers have used a modification of CRISPR-Cas9 gene editing to correct a disease-causing mutation in some patients with heart failure (it didn’t work for everyone). That extended their life expectancy from less than six months to over 18 years by significantly improving their ejection fraction — which is a measurement of how much blood the left ventricle pumps out with each contraction. Plus, gene editing is now being used to enhance cancer immunotherapy. In 2022, the Food and Drug Administration granted orphan drug designation to a gene- edited T-cell therapy for acute myeloid leukemia. Now imagine knocking out the gene that is responsible for producing the protein that causes most atherosclerosis. That trial is underway, since it already succeeded in preventing heart disease and stroke in animal models. And there’s more: The Nobel-prize winning discovery of four Yamanaka factors is being dubbed the Fountain of Youth. Using three of these factors, it is possible to reset 80- to 90-year-old human skin and eye cells so they look and function like those of a 24-year-old.
